Targeting strategy
Gene targeting strategy for B-hFSHR mice. The exon 2 of mouse Fshr gene that encodes the full-length protein was replaced by human FSHR exon 2 in B-hFSHR mice.
mRNA expression analysis
Strain specific analysis of FSHR gene expression in wild-type C57BL/6 mice and B-hFSHR mice by RT-PCR. Mouse Fshr mRNA was detectable in ovary and testis of wild-type C57BL/6 mice (+/+). Human FSHR mRNA was detectable only in homozygous B-hFSHR mice (H/H) but not in wild-type mice.
Protein expression analysis
Strain specific FSHR expression analysis in homozygous B-hFSHR mice by western blot. Ovary, testis were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hFSHR mice (H/H), and analyzed by western blot with anti-FSHR antibody. FSHR was detectable in wild type mice and homozygous B-hFSHR mice, as the antibody is crossly reactive with FSHR in human and mice. FSHR was not detectable in FSHR KO mice (-/-).
